Abstract: Охорона реліктових видів рослин є важливим напрямом збереження біорізноманіття України. До рідкісних високогірних видів Українських Карпат належать первоцвіт Галлера (Primula halleri J.F.Gmel.) та первоцвіт дрібний (Primula minima L.), занесені до Червоної книги України. Перспективним методом їх збереження є культура in vitro, що забезпечує отримання життєздатного рослинного матеріалу та ефективне розмноження за умов низької схожості насіння. Встановлено вплив еколого-географічних умов на вікову структуру та самопідтримання популяцій P. minima і P. halleri. Оптимізовано умови стерилізації насіння, ефективність якої становила 97–99%. Підібрано умови вегетативного розмноження in vitro: для P. minima – середовище МС/2 з 0,5 мг/л ОК, 0,5 мг/л ГК₃ та 0,05 мг/л Кін, для P. halleri – безгормональне середовище МС/2. Для мікроклонального розмноження оптимальними були середовища МС/2 з 0,2 мг/л ІОК, 0,7 мг/л ГК₃, 0,03 мг/л Кін і 0,01 мг/л БАП для P. minima та з 0,1 мг/л Кін і 0,05 мг/л ГК₃ для P. halleri. Розроблено схему адаптації рослин до умов in vivo та підтверджено можливість їх успішного вирощування у високогір'ї Українських Карпат, де приживання рослин становило 17 % для P. minima та 55 % для P. halleri.
The protection of relict plant species is an important direction in the preservation of Ukraine’s biodiversity. Rare high-altitude species of the Ukrainian Carpathians include Haller's primrose (Primula halleri J.F.Gmel.) and small primrose (Primula minima L.), listed in the Red Book of Ukraine. A promising method for their preservation is in vitro culture, which ensures the receipt of viable plant material and effective propagation under conditions of low seed germination. The influence of ecological and geographical conditions on the age structure and self-maintenance of P. minima and P. halleri populations was established. The conditions for seed sterilization were optimized, the efficiency of which was 97–99%. The conditions for vegetative propagation in vitro were selected: for P. minima – MS/2 medium with 0.5 mg/l OK, 0.5 mg/l GK₃ and 0.05 mg/l Kyn, for P. halleri – hormone-free MS/2 medium. For microclonal propagation, the optimal media were MS/2 with 0.2 mg/l IOK, 0.7 mg/l GK₃, 0.03 mg/l Kyn and 0.01 mg/l BAP for P. minima and with 0.1 mg/l Kyn and 0.05 mg/l GK₃ for P. halleri. A scheme for adapting plants to in vivo conditions was developed and the possibility of their successful cultivation in the highlands of the Ukrainian Carpathians was confirmed, where plant survival was 17% for P. minima and 55% for P. halleri.
